Glycine is the cheapest supplement with any sensible sleep research behind it. A kilogram of plain powder costs roughly what one tub of a fashionable sleep formula costs, and lasts a year. That is a good reason to look at the evidence more carefully than cheap things usually get looked at.
How much glycine for sleep?
Three grams, about 60 minutes before bed. That is what the Japanese studies from 2007 to 2012 used, and that is what is worth reproducing, because no data suggests more delivers more.

Glycine is a non-essential amino acid, present in the diet mainly through collagen and gelatin. A typical diet supplies 1.5 to 3 g a day, so supplementing 3 g roughly doubles the intake. You are not introducing a foreign substance, you are shifting the amount and, more importantly, the timing.
How does glycine work on sleep?
Glycine lowers core body temperature by dilating peripheral vessels, speeding up heat loss through hands and feet. Kawai’s 2015 study showed the effect depends on NMDA receptors in the suprachiasmatic nucleus, the body’s master clock.
That drop in core temperature is the physiological start signal for sleep. It is the same one exploited by a cool bedroom, a warm shower an hour before bed, and socks on cold feet.
This is the mechanism’s strength and also a practical warning: if your bedroom sits at 24 °C, glycine is fighting your thermostat. Temperature first, supplement second. The order is set out in the sleep protocol, and the temperature thresholds in sleep hygiene.
What did the glycine trials show?
In Inagawa’s 2007 study, volunteers with sleep complaints took 3 g of glycine before bed. They reported falling asleep faster and rated sleep quality higher on questionnaires, while polysomnography showed shortened latency to slow-wave sleep.
Bannai showed in 2012 that glycine improves subjective daytime performance in people whose sleep had been partially restricted. That is an important distinction: the trial measured how participants felt the next day, not sleep architecture.
Notice what was actually measured. Most of the strong results concern subjective ratings and latency rather than total sleep time or number of awakenings. Glycine looks like an agent that shortens sleep onset, not one that lengthens sleep.
What do the glycine trials not show?
Samples were small, in the teens, run mostly at a single centre and funded by an amino acid manufacturer. There is no large independent replication and no data on use beyond a few weeks. That is why I label this preliminary rather than proven.
There are also no trials in people with diagnosed chronic insomnia, and no head-to-head comparison against melatonin or against cognitive behavioural therapy for insomnia, which remains the first-line treatment with the strongest evidence.
Honestly, I do not know how much to believe these results. The mechanism is convincing and consistent with what is known about sleep thermoregulation. The sample sizes and the funding source argue for keeping expectations low. I take glycine on and off, and I cannot separate its effect from the fact that on those same evenings I go to bed at a consistent time.
Where glycine got popular
Glycine reached awareness outside Japan roughly when collagen and single amino acids became fashionable. It was helped by being cheap, having an understandable mechanism, and not needing to explain itself on safety.
What helped it also hurt it. Around a cheap compound with a simple mechanism, claims grew much faster than the research: about recovery, about glutathione levels, about longevity. The last of those rests on animal models and on glycine’s role as a substrate for glutathione synthesis, which is a mechanism rather than a clinical result.
I stick here to what was measured in humans: sleep latency and next-day wellbeing. The rest may turn out to be true, but for now it is a hypothesis.
Glycine versus melatonin, magnesium and CBT-I

That table carries an uncomfortable conclusion. The strongest evidence for insomnia belongs to something that is not a supplement and costs a few hundred pounds: cognitive behavioural therapy for insomnia. No shop sells it, which is why you will not see it in “best sleep supplements” rankings.
A common melatonin error: the 3 to 5 mg doses sold in most shops are several times higher than the doses used in phase-shifting research. More is not better here, and excess brings morning grogginess.
Who probably will not benefit from glycine?
People who fall asleep in 10 minutes and wake rested. Glycine acts on latency, and if your latency is already short there is nothing to shorten.
People whose problem is waking in the middle of the night rather than falling asleep. Glycine trials show no improvement on that measure, and 3 am awakenings more often trace back to alcohol, apnoea or anxiety than to thermoregulation.

Which form of glycine should you buy and what does it cost?
Buy plain glycine powder. It tastes sweet, dissolves in water and needs no encapsulation. A kilogram runs about £10 to £18, which at 3 g a day gives roughly 330 doses.
Capsules are more convenient and noticeably more expensive, and at a 3 g dose mean swallowing several of them. Collagen delivers glycine alongside the rest of its amino acids, but at 20 to 22% content you need around 14 g of collagen for 3 g of glycine.
What are the most common mistakes in use?
Taking it right as you get into bed. The thermoregulatory effect needs time, which is why trials used an hour of lead time rather than five minutes.
Pairing it with a protein-heavy dinner in the hope of amplifying the effect. A large meal raises postprandial thermogenesis, which does the exact opposite of what you want.
Judging after two nights. Night-to-night variation in latency can exceed the expected effect, so two nights tell you nothing. The minimum is two weeks with a log, as set out in the sleep protocol.
Checklist before the first dose
Check your bedroom temperature. If it is above 20 degrees, start there, because the supplement will be working against the environment.
Check whether your problem is falling asleep or staying asleep. Glycine only has data on the former.
Check your medication list for clozapine, and consult a doctor if you have kidney disease.
Plan two weeks with no other changes, and prepare somewhere to log your latency. Without that last point the whole test is worthless, because after two weeks you will remember the last three nights rather than all fourteen.
How do you test glycine on yourself?
Two weeks without, two weeks with, everything else unchanged. Log sleep latency, meaning your estimate of the time from lights out to falling asleep.
If the difference comes out under 10 minutes, that is noise rather than effect. Natural night-to-night variation in latency is larger than the expected effect of the supplement, so a single good night tells you nothing.

How much glycine should I take for sleep?
Trials used three grams taken about 60 minutes before bed. The dose is reproducible and cheap: 3 g of plain glycine powder costs pennies. Higher doses have not been studied for sleep and there is no reason to use them.
Is glycine safe?
At 3 g per day the safety profile is good, with mild stomach upset the most common side effect. It needs care alongside clozapine, because glycine may reduce that drug's effect. With kidney disease, make the decision with your doctor.
Glycine or magnesium for sleep?
They work through different mechanisms: glycine lowers core body temperature, magnesium acts on GABA signalling and muscle tone. If you test both, test them one at a time for two weeks each, or you will not know which one worked.
How quickly does glycine start working?
The effect on sleep latency appears from the first night, because the mechanism is thermoregulatory rather than cumulative. If a week of nightly use makes no difference to how long you take to fall asleep, it probably will not work for you.
Does glycine from collagen work the same way?
Collagen is about 20 to 22% glycine, so 10 g of collagen delivers roughly 2 g of glycine. That approaches the trial dose, but it also delivers the other amino acids and costs several times more than plain glycine.
